Alu pcr lab
In this lab, our group of Leo, Angus, Elise, Aidan, and Ryan attempted to find specific Alu in our DNA sample. We partook in the lab for a couple days but in the end we found that the work had payed off; a little.
Purpose: Our purpose was to use PCR (Polymerase Chain Reaction) to amplify a dimorphic Alu insert repeat.
Hypothesis: The hypothesis for our group was that we would be able to find out about our Alu frequency.
Materials:

Pipet
Micropippets
Racks
Microfuge tubes
Microcentrifuge
Heat Box
Thermal Cycler
(Reagents:)
Cheek cells
Chelex
Master mix: nucleotides, DNA polymerase
Primer Mix
DNA Strain Solution
TBE Buffer, Agarose Gel
Procedure:
1. Swoosh a saline solution in our mouths for 30 seconds to get cheek cells
2. Expel saline into a cup and mix the cells.
3. Microcentrifuge the saline solution
4. Pour the supernatant into the access supernatant without pouring the cell pellet
5. Extract 100uL of the saline covering the cells WITHOUT EXTRACTION THE CELLS
6. Withdraw 50uL of the cell suspension(chelex) and put it with the cells
7. Heat the mixture to 99*C for 10 minutes
8. Centrifuge the mixture after taking it out
9. Withdraw 50uL from the DNA WITHOUT transfering and Chelex beads
10. Refrigerate overnight
11. Pipet 20uL of Master Mix and 20uL of the primer mix
12. Pipet 10uL of the extracted DNA into the PCR tube
13. Microfuge the tube for 10 seconds
14. Add 5uL of loading dye into your PCR tube
